abstract | FIELD: molecular biology, biochemistry. SUBSTANCE: invention proposes method for enzymatic synthesis of DNA by carrying out polymerase chain reaction using thermostable DNA polymerase. Synthesis is carried out in the presence of saturated solution of difficultly soluble magnesium salt magnesium oxalate. The use of magnesium oxalate allows providing concentration of magnesium ions that is optimal for DNA synthesis being at temperature above 50-55 C only that inhibits initiation of synthesis of nonspecific DNA sequences. This method provides high specificity of DNA synthesis. Invention can be used for synthesis, detection and identification of DNA in biological samples.
