abstract | FIELD: analytical methods in medicine. SUBSTANCE: method comprises sampling of body fluid, isolating hydroxyproline therefrom, determining concentration of free and protein-bound hydroxyproline, estimating degradation of collagen and its biosynthesis process from, respectively, increase in concentration of free and protein-bound hydroxyproline. Distinguishing feature of invention consists in that above-mentioned body fuel is saliva from a person to be examined in amount 4 ml. It is mixed with 16 ml ethanol, mixture is stirred and centrifuged for 15 min at 3000 rpm, after which protein- bound hydroxyproline is isolated on chromatographic column with ion-exchange resin (KRS-8p) 3 mm in diameter and 6 mm of height of its filled part at neutralizate motion velocity 0.2 ml/min. Hydroxyproline is eluted with 1 ml 1 N hydrochloric acid, after which hydrochloric eluate is neutralized with 5 N NaOH. EFFECT: improved quality of analysis and eliminated trauma risk. |
