abstract | FIELD: medicine, cytomorphology. SUBSTANCE: method deals with cytological analysis of mono-layer cultures of human and animal cells. Fixed cells should be poorly dyed with the main fuchsin and studied with a light microscope with the help of phase-contrast device in orange-yellow-green area of the spectrum (within 5000-6300 wave length range), by differentiating certain cellular structures by morphology and color. Moreover, mitochondria produce black color, granules of lysosomal type - from yellow to yellowish- brown color, cisterns of endoplasmatic net - from yellow to yellowish-green color, lamellar complex - brown color, nuclear membrane (by contour) and nucleolus - from dark-blue to black color, chromatin net - from brown to reddish-brown color; morphological formations of cytoplasmatic membrane in marginal area (contours of lamellipodia, phylopodia, microvilli and lamelloplasma) - from dark-green to black color. The method provides the opportunity to obtain a multi-color picture, moreover some organelles and cellular structures obtain their own color steadily that simplifies the possibility of their differentiation.
