abstract FIELD: biology, molecular biology, medicine. SUBSTANCE: invention relates to field of regulation of cell growth. Synthetic mutant genes Rb and p53 are mutated by alternation of nucleotides encoding amino acids in conservative homologous regions for nucleotides encoding nonconservative amino acids. Amino acid sequences are given in description. Mutated regions comprise the same amount, more or less amino acids as compared with the region of wild type. Synthetic mutant gene Rb is mutated by alternation of nucleotide sequence that encodes amino acids in phosphorylation sites of protein Rb. Plasmid is constructed on the basis of plasmid Rb HuBa cpr-1-neo-PQ with incorporation of mutant gene Rb or p53 in it. Cellular proliferation is inhibited by insertion of these plasmids into nonmalignant proliferating cell. Retinoblastoma protein is encoded by synthetic mutant gene Rb. Retinoblastoma protein p53 is encoded by synthetic mutant gene p53. Retinoblastoma protein p53 is inserted into cell for induction of apoptosis. Proliferation of tumor cell with mutation of cyclin protein is inhibited by insertion of retinoblastoma protein p53 into cell. Invention ensures to treat pathological cellular-proliferative states.
