FIELD: organic chemistry, nucleosides, pharmacy. SUBSTANCE: invention relates to novel antiviral derivatives of 3'-azido-3'-deoxythymidine 5'-H- phosphonate of the general formula (I): where R means isopropyl, neopentyl or cyclohexyl group and to pharmaceutical compositions comprising thereof. Compounds of the formula (I) are synthesized by interaction of alcohol ROH with phosphorus trichloride and the following addition of 3'-azido-3'-deoxy-thymidine 5'-H- phosphonate to reaction mixture or by interaction of 3'-azido-3'-deoxythymidine 5'-H-phosphonate with alcohol ROH in the presence of pivaloyl chloride as an activating agent. Compounds of the formula (I) show the decreased hydrophoby and enhanced penetration across cellular membranes. Pharmaceutical compositions containing compounds of the formula (I) taken in effective therapeutic dose and pharmaceutically acceptable carrier show an antiviral activity, among them, against human immunodeficiency virus (HIV).
