abstract | FIELD: biotechnology, biochemistry, preparative enzymology. SUBSTANCE: invention relates to methods of preparing the pancreatic ribonuclease and can be used in medicinal, chemical and microbiological industry. Method of preparing the pancreatic ribonuclease involves stages of tissue grinding, extraction of ribonuclease, separation of suspended particles by centrifugation, precipitation of ribonuclease, its purification from impurities, washing out with ethyl alcohol and drying. After separation of suspended particles by centrifugation or filtration extract is subjected for ultra- concentrating on membrane UPM-450 with recovery of acid permeate to the extraction stage. Precipitation of enzyme is carried out at its isoelectric point followed by thermoinactivation of proteins eliciting protease and DNAse activity. Method provides the increase of the preparation yield from 0.1% to 0.6% of pancreas dry mass, i. e. by 6 times. EFFECT: improved method of enzyme preparing, increased yield. |
