titleOfInvention Strain of bacterium citrobacter diversus n 256 showing pathogenicity factors complex
abstract FIELD: microbiology. SUBSTANCE: invention relates to preparing thermolabile enterotoxin and anatoxin from Citrobacter diversus in vaccines producing. Strain is isolated from feces of patient with acute intestine infection and deposited in Collection of the State Scientific- -Research Institute of standardization and control of medicinal biological preparations at number 256. Citrobacter diversus GISK N 256 shows the following traits: strain grows on usual universal solid nutrient media forming typical isolated colonies. In beef-extract broth strain causes the uniform turbidity, precipitate formation and film formation on surface. Subplantar (model of paw edema) administration in mice (weight 15-16 g) of 0.05 ml of centrifugate of 20 h broth culture causes paw edema with difference above 120 mg. Intranasal administration of 80 mln of bacterial cells causes the death of 95% of mice-male (weight 15-16 g) for 1.5 h. Intracutaneous administration of 300 mln of bacterial cells of 20 h broth culture causes skin necrosis in rabbits (diameter above 3.5 cm). In experiment with rabbit intestine loop administration of the strain in the dose 500 mln of bacterial cells gives the positive effect (V/L = 1.2). Centrifugate of 20 h broth culture heated at 56 C for 45 min causes paw edema in mice (below 30 mg) but does not cause skin necrosis in rabbits. Heated 20 h culture (at 100 C) does not cause fluid accumulation in rabbit intestine isolated loop (V/L = 0.3). Conditions and medium composition for multiplication: Hottinger's broth (pH 7.2-7.4), 1.5% beef-extract broth at 37 C for 18-24 h. EFFECT: increased toxin-producing capacity of strain of bacterium Citrobacter diversus GISK N 256.
