abstract | FIELD: medicinal microbiology. SUBSTANCE: method involves preparing a series of 10-fold dilutions of analyzed material in liquid selective nutrient enrichment medium at 39 C, seeding from dilutions on surface of solid selective nutrient medium, culturing seedings for 24-48 h at increased content of carbon dioxide gas (5%) and decreased oxygen concentration (16%) in culturing atmosphere. Liquid selective enrichment nutrient medium has the following composition, g/l: defatted milk liquid hydrolyzate, 225.0 ± 25.0; yeast concentrated autolyzate, 110.0 ± 10.0; agar, 0.8; distilled water, up to 1 l; 20% solution of sodium hydroxide, up to medium pH 5.4 ± 0.1. Then lactobacilli are isolated in liquid selective nutrient medium of the following composition, g/l: defatted milk liquid hydrolyzate, 225.0 ± 25.0; yeast concentrated autolyzate, 110.0 ± 10.0; agar, 22.5 ± 2.5; distilled water, up to 1 l; 20% solution of sodium hydroxide, up to medium pH 5.4 ± 0.1. Invention can be used for selective isolation of bacterium of genus Lactobacillus from clinical material (feces, vaginal secret). EFFECT: improved method of isolation, improved diagnosis of gynecological diseases. |
