abstract FIELD: molecular biology, genetic engineering. SUBSTANCE: invention relates to development of a method of activation of transcriptionally silent gene in genome of eucaryotic cellular line and an expression of product encoding by the indicated gene. Method involves an insertion of DNA-construct into genome by homological recombination. DNA-construct has regulatory DNA segment that is able to stimulate gene expression and catching DNA segment that shows homology with respect to genome site at limits or proximally to the indicated gene and bound with it functionally. After insertion of DNA-construct its regulatory segment is bound functionally with an active end gene. Then method involves culturing a cellular line at conditions exhibiting benefit for expression of gene product and collection of this product. Method can be used for modification of expression properties of any endogenic gene of this cellular line. EFFECT: improved method of an activation and expression. 16 dwg, 1 ex
