titleOfInvention Method of an express-diagnosis of diphtherial infection
abstract FIELD: medicine, diagnosis of infection diseases. SUBSTANCE: method involves detection of diphtherial toxin by immuno-enzyme analysis using monoclonal antibodies. Reaction is carried on mouth-throat mucosa epitheliocytes that are washed out preliminary with phosphate-saline buffer, pH = 7.2-7.4, in combination with Tween-20. Mouth-throat mucosa bioptate is applied by endoscope brush on glass, fixed in 15-20% sucrose and phosphate-saline buffer solution (pH = 7.2-7.4) with 1% bovine serum albumin solution. Then monoclonal antibodies raised to COOH-terminal site of B-fragment of toxin molecule are applied followed by application of polyclonal antibodies with peroxidase and substrate mixture. Infection is diagnosed in the case of precipitation of antibodies of red-brown color. Method can be used for early diagnosis of infectious diseases.
