abstract FIELD: medicine, in particular, experimental pathology and immunology for investigation of cellular differentiation and transformation in inflammation nidus, as well investigation of interaction between cells in nidus of immune and non-immune granuloma inflammation, development of methods for drug treatment of granuloma decease. SUBSTANCE: method involves long-term monolayer culturing of macrophages and lymphocytes on glass. After joint culturing of macrophages and lymphocytes, granulocytes and abdomen wash fat cells, method involves removal of carrier with non-adhesive cells, return of non-adhesive macrophages and lymphocytes to culture of macrophages in fresh maintenance medium in which zymosan granules which are loaded with acid fuchsin are introduced. Mixed culture of macrophages and lymphocytes is incubated at 37 C in wet air with serum of cow embryos, penicillin and streptomycin. This operation is repeated every 3 days simultaneously with return of non- adhesive lymphocytes to culture of macrophages. Transient types of transformed macrophages, giant cells with multiple nuclei, clusters of epithelial cells are detected in culture of macrophages at different time intervals using optical microscopes.
