FIELD: biotechnology. SUBSTANCE: invention relates to methods of proteins splitting using IgA-proteases, in part, to methods of production of recombinant proteins or polypeptides in procaryotic organisms with possibility the following removal of N-terminal sequence. Recombinant granulocyte-colony-stimulating factor (G-CSF) with additional methionine residue at N-terminus is prepared by splitting the fused protein with IgA-protease. G-CSF is expressed in E. coli cells and this protein has recognition site located between N-terminal methionine and amino acid sequence of G-CSF. EFFECT: improved method of protein splitting. |
