titleOfInvention Method of histamine assay in sputum
abstract FIELD: medicine. SUBSTANCE: sputum sample is homogenized in equal volume of trichloroacetic acid and after centrifugation supernatant is isolated. Alkali solution and a mixture butanol-chloroform were added to supernatant to transfer histamine from aqueous phase to organic one. The centrifugation is carried out to separate these phases. Then organic phase containing dissolved histamine is separated from an aqueous phase and acid is added to transfer histamine to an aqueous phase followed by centrifugation. An aqueous phase is separated and ortho-phthalic aldehyde is added to an aqueous phase. Prepared condensate of histamine and ortho-phthalic aldehyde is stabilized by ortho-phosphoric acid addition and its concentration is measured by fluorimetry. EFFECT: improved method of histamine assay.
