titleOfInvention | Method of preparing african equine plague virus antigen |
abstract | FIELD: virology. SUBSTANCE: african equine plague virus is grown in cell culture or organ animal tissue. Virus-containing suspension is frozen once at -40 C, thawed out at the room temperature and cleared by centrifugation at 3000 rev/min for 20 min. Supernatant is controlled for activity and specificity for reaction of long boudiny of complement. Then virus is inactivated by methylenebismorpholine addition at the final concentration 0.1-0.2% for 48-72 hr at 36-38 C at periodic stirring. After inactivation antigen with titer in reaction of long boudiny of complement 1:8 is concentrated with 4% polyethylene glycol of molecular mass 6000-20000 Da by 35-100 fold and used for donor immunization of specific sera or as diagnostic antigen after preserving with sodium azide 1:5000 or lyophilic drying.
