titleOfInvention IMPROVED METHODS FOR ISOLATING RECOBINANT PROTEINS
abstract 1. A method for producing a recombinant protein, comprising (a) culturing a prokaryotic host cell, where the prokaryotic host cell is transformed with a nucleic acid encoding a recombinant protein, and (b) isolating the recombinant protein under conditions where the level of dissolved oxygen (dO) exceeds 0% and (c) purification of the recombinant protein to a filtered storage preparation (FBS), where the filtered preparation does not contain detectable amounts of a recombinant protein adduct and 1,4-dihydroxy-2-naphthoate (DHNA) according to ionoo transfer chromatography (IEC) at 310 nm. 2. The method of claim 1, wherein the analytical analysis is HPLC, RP HPLC, HIC HPLC, NMR, mass spectrometry or UV spectroscopy. The method of claim 1, wherein the dO level is continuously maintained at a level of more than 0% in the isolation step (b). The method of claim 3, wherein the isolation comprises a homogenization step. The method of claim 4, wherein the dO level is maintained at a level of from about 30% to about 75% prior to homogenization. The method of claim 4, wherein the dO level is maintained at a level of more than 75% until homogenized. The method of claim 4, wherein the dO level is maintained at about 50% after homogenization. The method of claim 5, wherein the dO level is maintained at about 50% after homogenization. A method according to claim 4, wherein the dO level is maintained at a level of more than 50% after homogenization. The method of claim 5, wherein the dO level is maintained at a level of more than 50% after homogenization. The method of claim 5, wherein the dO level is maintained for a period equal to or greater than 1.5 hours. The method of claim 6, wherein the dO level is maintained for a period equal to or greater than 2 hours. The method according to claim 1, wherein the dO level is maintained using air injected into the space above the medium or bubbler
