titleOfInvention | The process of isolating the target oligonucleotide from the mixture |
abstract | 1. A method of isolating a target oligonucleotide from a mixture of a target oligonucleotide and one or more impurities, comprising: providing a two-phase liquid-liquid chromatographic system mobile phase - a stationary phase containing a first liquid mobile phase and a liquid stationary phase containing at least one exchanger substance which removably binds to the target oligonucleotide; causing the first liquid mobile phase to transfer the target oligonucleotide in a stream relative to and in contact with the liquid stationary phase in the column of the liquid-liquid chromatography apparatus, so that the target oligonucleotide becomes bound to the exchanger in the liquid stationary phase; then the target oligonucleotide is displaced from the liquid stationary phase into the second liquid mobile phase, which flows relatively and in contact with the stationary phase through the column by a propellant capable of displacing a target oligonucleotide from a liquid stationary phase into a second mobile phase. 2. The method of claim 1, comprising the steps of: (1) providing a first liquid phase containing a target oligonucleotide and one or more impurities in a solution, and a second liquid phase containing an exchanger substance that is removably bound to the target oligonucleotide, wherein the first and second the liquid phases form two separate phases upon contact with each other; (2) the introduction of the second liquid phase into the centrifugal distribution chromatography apparatus as a stationary liquid phase; (3) the introduction of the first liquid phase containing the target oligonucleotide and one and whether there are more impurities in solution in a centrifugal distributor |
