titleOfInvention METHOD FOR PRODUCING POLYPLOID MEGACARIOCYTES AND THROMBOCYTES
abstract 1. A method of obtaining polyploid megakaryocytes, including the forced expression of the apoptosis suppressor gene in megakaryocytes that have not undergone polyploidization, and culturing these cells. The method of claim 1, wherein the apoptosis suppressor genome is the BCL-XL gene. The method according to claim 1, in which at the stage of cultivation inhibit the expression or function of the p53.4 gene product. The method according to claim 1, wherein at the culturing stage, megakaryocytes that have not undergone polyploidization are subjected to at least one of the following treatments (a) to (c) :( a) treatment with an actomyosin complex function inhibitor; (b) treatment with a ROCK inhibitor; and (c) treatment with an HDAC inhibitor. 5. The method of claim 4, wherein the ROCK inhibitor is Y27632; the HDAC inhibitor is valproic acid; and the actomyosin complex function inhibitor is blebbatin. 6. The method according to claim 1, in which the stage of cultivation is carried out at a temperature above 37 ° C. The method according to claim 1, in which megakaryocytes that have not undergone polyploidization are obtained by forced expression of an oncogen and any of the following genes (i) - (iii) in cells at any stage of differentiation from hematopoietic progenitor cells to megakaryocytes that have not undergone polyploidization :( i) a gene that suppresses the expression of the p16 gene or p19 gene; (ii) a gene that suppresses the expression of the Ink4a / Arf gene; and (iii) a polycomb gene; cultivation and proliferation of the obtained cells. 8. The method of claim 7, wherein the c-MYC gene is used as the oncogene, and BMI1.9 is used as any of the genes (i) to (iii). The method of claim 7, wherein the hematopoietic progenitor cells are derived from cells selected from the group consisting of IPA cells, ES cells, hematopoietic stem cells
