titleOfInvention | OPTIONS OF PLASMINOGEN AND PLASMINE |
abstract | 1. An isolated variant of plasminogen, characterized in that: (i) it contains in its catalytic domain lysine or arginine in the internal position P and a mutation of at least one amino acid in position [P +/- n], where n is 1, 2, 3, 4 or 5, and where the amino acid at the position [P +/- n] is not lysine and is not arginine; and (ii) the presence of a mutation from (i) reduces the degree of autoproteolytic degradation of a plasmin variant derived from said plasminogen variant compared to the degree of autoproteolytic degradation of wild-type plasmin, which is, for example, determined by chromogenic analysis or analysis of biological activity to a substrate. . A plasminogen variant according to claim 1, comprising in its catalytic domain a mutation of at least two internal amino acids: - at the positions [P +/- n] and [P +/- n ']; where n and n 'are 1, 2, 3, 4 or 5; where the amino acids at the positions [P +/- n] and [P +/- n '] are not lysine and are not arginine; and where n and n 'are different from each other if both are either added or subtracted from P; or - in the provisions of [P +/- n] and [P '+/- n]; where P 'is the larger value of P and P'; where the amino acid at position P 'is lysine or arginine; where n is 1, 2, 3, 4 or 5; where the amino acid at position [P '+/- n] is not lysine and is not arginine; and where, if any, positions overlapping between [P + n] and [P'-n] are excluded; and where the presence of a mutation of the at least two internal amino acids reduces the degree of autoproteolytic degradation of the plasmin variant obtained from the plasminogen variant, compared with the degree of autoproteolytic decomposition of wild-type plasmin, which, for example, was determined using chromogenic analysis or |
