abstract 1. A chromatographic separation method for isolating a polyunsaturated fatty acid (PUFA) product from an initial mixture, which comprises introducing the initial mixture into a chromatographic unit with a pseudo-moving or true moving layer, having many associated chromatographic columns containing, as an eluent, aqueous alcohol, where the installation has many zones, including at least a first zone and a second zone, each zone having an extract stream and a raffinate stream, from which liquid can be taken from a plurality of coupled chromatographic columns, and where (a) the raffinate stream containing the PUFA product together with the more polar components is selected from the column in the first zone and introduced into the non-adjacent column in the second zone and / or (b) the extract stream containing the PUFA product together with less polar components, is selected from the column in the second zone and introduced into the non-adjacent column in the first zone, and the specified PUFA product is separated from the other components of the initial mixture in each zone. 2. The method of claim 1, wherein a portion of one or more of the extract stream from the first zone, the raffinate stream from the first zone, the extract stream from the second zone, and the raffinate stream from the second zone recycles back to the same zone, typically to an adjacent column in that same zone. 3. The chromatographic separation method according to claim 1, where (a) the water-alcohol eluent present in each zone has a different ratio of water: alcohol; and / or (b) the rate at which the fluid drawn through the extract and raffinate streams in each zone recirculates back to the same zone is adjusted so that the PUFA product
