abstract 1. A method of measuring the activity of a cell-mediated immune response in a subject, where the method includes contacting a sample of lymphocytes from the subject with one or more agents that enhance the acquired and innate immunity systems, and measuring the presence or increase of the level of the immune effector molecule from immune cells, where the presence or the level of an immune effector molecule indicates the level of cell-mediated reactivity in a subject. 2. The method according to claim 1, the subject is a human. The method of claim 1, wherein the sample is undiluted whole blood. The method according to claim 3, where the sample is whole blood, which is from 10% to 100% of the volume of the analyzed sample. The method according to claim 4, wherein the whole blood is from about 50% to 100% of the volume of the analyzed sample. The method according to claim 5, wherein the whole blood is from about 80% to 100% of the volume of the analyzed sample. The method according to claim 1, where the sample is represented by a small blood volume from 1 μl to 1000 μl. The method of claim 1, wherein the lymphocyte source is further incubated with the antigen. The method of claim 3, wherein the whole blood is collected in tubes containing heparin. The method of claim 1, wherein the agent that enhances the acquired immune response is a T cell receptor agonist. The method of claim 10, wherein the agent is selected from a CpG oligonucleotide, antibodies to a complex of a T-cell receptor, phytohemagglutinin and anti-CD3 antibody. The method of claim 1, wherein the agent that enhances the innate immunity system is a Toll-like receptor agonist (TLR).
