abstract | 1. A protein capable of binding the extradomain B (ED-B) of fibronectin, including a modified heterodimeric ubiquitin protein, in which two monomeric ubiquitin units are connected to each other in a head-to-tail arrangement, characterized in that each monomer of said dimeric protein is different modified by substitutions of at least 6 amino acids at positions 2, 4, 6, 8, 62, 63, 64, 65, 66 and 68 of SEQ ID NO: 1, said substitutions including: (1) in the first monomeric substitution unit of at least least in amino acid positions 6, 8, 63, 64, 65 and 66; and the second monomeric substitution unit at least at amino acid positions 6, 8, 62, 63, 64, 65 and 66; optionally an additional 2, or (2) in the first monomeric substitution unit at least at amino acid positions 2, 4, 6, 62, 63, 64, 65, and 66; and the second monomeric substitution unit at least at amino acid positions 6, 8, 62, 63, 64, 65 and 66; optionally, additionally 2, and optionally other modifications, preferably substitutions of other amino acids, wherein said modified monomeric ubiquitin unit has an amino acid identity with SEQ ID NO: 1, at least one from the group of 80%, at least 85%, and at least 90%, and wherein said protein has a specific binding affinity for said fibronectin ED-B domain with kD = 10-10M and exhibits monovalent binding activity with said fibronectin extradomain B (ED-B). 2.
