titleOfInvention | METHOD AND KIT FOR DETECTION OF MICROORGANISMS |
abstract | 1. A method for detecting viable cells of a microorganism in a test sample when distinguishing between viable cells and dead cells or damaged cells, which includes the steps: a) adding to the test sample an agent capable of covalently binding to DNA or RNA when irradiated with light with a wavelength of 350 nm or more up to 700 nm; b) irradiating the test sample to which the agent is added with light with a wavelength of 350 nm to 700 nm; the action of a substance that inhibits the amplification of nucleic acids, without the release of nucleic acids from cells; and d) analysis of the amplification products. 2. The method according to claim 1, wherein the amplification of the target region is carried out in the cells of the microorganism. The method according to claim 1 or 2, wherein in step c) the amplification of the target region is carried out in the presence of one or more classes selected from a surfactant, a magnesium salt and an organic acid salt or a phosphoric acid salt. Method according to claim 1 or 2, wherein before step c), steps a) and b) are repeated. The method according to claim 1 or 2, wherein before step a), the following step e): e) treatment of the test sample with an enzyme with the activity of destroying cells other than cells of a microorganism, colloidal particles of protein, lipids or saccharides present in the test sample.6 . The method of claim 5, wherein the enzyme is selected from a protein degrading enzyme, a lipid degrading enzyme, and a saccharide degrading enzyme. The method according to claim 1 or 2, where the test sample is any of the food product, bio |
