abstract 1. A method for detecting activity of endopeptidases with a changed focus, comprising the steps of: a. treating a cell from a stable cell line with a sample containing an altered targeting endopeptidase, said cell from a stable cell line being sensitive to the activity of said endopeptidase with an altered focus at an endopeptidase concentration with an altered focus of about 500 pM or lower; b. isolating from the treated cell the SNAP-25 component containing the SNAP-25 cleavage product, the carboxyl end of which corresponds to the glutamine of the cut bond at the BoNT / A toxin cleavage site; c. contacting the SNAP-25 component with an anti-SNAP-25 antibody immobilized on a solid phase support, while the anti-SNAP-25 antibody binds to an epitope whose carboxyl end corresponds to the glutamine of the cut bond in the BoNT / A toxin cleavage site from the SNAP cleavage product -25, anti-SNAP-25 antibody is characterized by an association rate constant for an epitope of SNAP-25 that does not contain the carboxy-terminal glutamine of the cleaved bond at the BoNT / A toxin cleavage site, from the SNAP-25 cleavage product of less than 1 × 10 Ms; and the anti-SNAP-25 antibody is characterized by an equilibrium dissociation constant for the epitope of less than 0.450 nM; g. detecting the presence of an antibody-antigen complex comprising an anti-SNAP-25 antibody and an SNAP-25 cleavage product, the carboxyl end of which corresponds to the glutamine of the cleaved bond at the BoNT / A toxin cleavage site; wherein detection with the indicated antibody-antigen complex is an indicator of endopeptidase activity with a changed focus
