titleOfInvention Modified Spider Silk Protein
abstract 1. A method for modifying spider silk protein, including the stages:! a) obtaining a nucleic acid sequence encoding a spider silk protein, which contains one module of SEQ ID NO: 1 or a variant thereof, where the variant contains 1 amino acid substitution, 1-2 amino acid deletions and / or 1-2 amino acid insertions, and which does not contain lysine or cysteine residues; ! b) replacing a nucleic acid sequence encoding serine and / or glycine in said module of said spider silk protein with a nucleic acid sequence encoding lysine and / or cysteine, and / or adding to said sequence a nucleic acid sequence encoding an amino terminal TAG according to SEQ ID NO : 20-24, containing lysine or cysteine along with other amino acids, and / or carboxy terminal TAG according to SEQ ID NO: 25-28, containing lysine or cysteine along with other amino acids; ! c) expressing the modified nucleic acid sequence obtained in (b) in a suitable host; and! d) isolating the expressed modified spider silk protein. ! 2. The method of claim 1, further comprising linking other substances to said lysine and / or cysteine molecule in the modified spider silk protein. ! 3. The method of claim 2, wherein the substance to be bound to said lysine and / or cysteine molecule is a marker molecule. ! 4. The method of claim 1, wherein the spider silk protein encoded by the nucleic acid sequence obtained in (a) comprises AQ or QAQ. ! 5. The method of claim 4, wherein the spider silk protein encoded by the nucleic acid sequence obtained in (a) comprises (AQ) 12, (AQ) 24, (QAQ) 8, or (QAQ) 16. ! 6.
