abstract 1. Method for selective enrichment and/or separation of N-terminal fragments of polypeptides and/or peptides in a sample, including: ! (a) double chemical labeling of proteins, polypeptides and/or peptides included in at least the first sample; ! (b) double chemical labeling of proteins, polypeptides and/or peptides included in at least the second sample; !(c) combining labeled proteins, polypeptides and/or peptides of steps (a) and (b); ! (d) hydrolysis of labeled proteins, polypeptides and/or peptides with the formation of their fragments; ! (e) fractionating said fragments; ! (f) performing a methylation reaction with said moieties; ! (g) re-fractionation of these fragments; ! (h) comparison of fractionated samples obtained in steps (e) and (g); and ! (i) separating the N-terminal fragments of said polypeptides and/or label-bearing peptides based on the results obtained in step (g), ! where double chemical labeling in steps (a) and (b) includes isotopic and isobaric labeling. ! 2. The method of claim 1, wherein at least one chemical label used in steps (a) and (b) is configured to label the primary amino groups of said polypeptides and/or peptides. ! 3. The method of claim 2, wherein the primary amino groups are the N-terminus of said polypeptides and/or peptides. ! 4. The method according to any one of claims 1 to 3, where at least two different isotopic labels are used in steps (a) and (b). ! 5. The method of claim 1, wherein the isotopic and/or isobaric labels used in step (a) and/or (b) further comprise an affinity group. ! 6.
