titleOfInvention A NEW METHOD FOR SEPARATING AND DETERMINING THE VIRAL LOAD IN THE SAMPLE OF PANCREATIN
abstract 1. A method for separating the viral load from a pancreatin sample, which consists in carrying out the following stages, at which: ! a) prepare a liquid pancreatin test sample suitable for centrifugation from the pancreatin sample without changing its viral load; ! b) subjecting at least one certain part of the prototype pancreatin obtained in step a) of the method to at least one low-speed centrifugation under conditions in which viruses characterized by sedimentation constants ≥120 S do not form debris; ! c) discard all solid deposits that are optionally formed in step b) of the method during low-speed centrifugation, and save the supernatant of the pancreatin prototype; ! d) subjecting at least one certain portion of the supernatant of the prototype pancreatin obtained in step c) of the method to a first ultracentrifugation in a stepped gradient medium, where the duration of the first ultracentrifugation and the relative centrifugal force for the first centrifugation are chosen so that the viral load is transported quantitatively from of the supernatant of the prototype pancreatin into the first target fraction located above the boundary layer or in the boundary layer between the lowest concentration gradient component located on top and the next higher concentration gradient component located below, and the first target fraction is quantitatively isolated, optionally including any debris present after centrifugation, which contains the viral load, from the supernatant
