titleOfInvention INTERFERON ALPHA 2B, MODIFIED BY POLYETHYLENE GLYCOL, PRODUCTION AND USE OF THE PRODUCT
abstract 1. Pegylated interferon-α2b (IFN-α2b) having the following structural formula, obtained by combining IFN-α2b with a Y-shaped branched polyethylene glycol (YPEG):! ! where! Ra and Pb are the same or different polyethylene glycols (PEG); ! j is an integer from 1 to 12; ! Ri-H is a substituted or unsubstituted C1-C12 alkyl group, substituted aryl, aralkyl or heteroalkyl; and! X1 and X2 separately and independently from each other represent a connecting group, where X1 is (CH2) n, and X2 is selected from the group consisting of (CH2) n, (CH2) nOCO, (CH2) nNHCO and (CH2) nCO where n is an integer from 1 to 10.! 2. The pegylated IFN-α2b according to claim 1, having the following structural formula! ! where R and R 'individually and independently from each other represent a C1-C4 alkyl group, preferably methyl; ! j is an integer from 1 to 12; ! m and m 'denote the degree of polymerization and may be any integers; ! m + m 'is preferably from 600 to 1500, and the average total molecular weight of PEG is preferably from about 10,000 to 60,000 Daltons, and most preferably about 40,000 Daltons. ! 3. The pegylated IFN-α2b according to claim 1 or 2, characterized in that PEG is linked to IFN-α2b via an amide bond formed by the α-amino group of the N-terminal amino acid IFN-α2b or the ε-amino group of the side chain of the Lys residue IFN-α2b at position 31, 49, 70, 83, 112, 121, 131, 133, 134 or 164 of the amino acid sequence designated as SEQ ID No.1, preferably at position 134 of SEQ ID No.1. ! 4. The pegylated IFN-α2b according to claim 3, characterized in that IFN-α2b is extracted from a natural source or obtained by recombinant biotechnology and it is preferably human IFN-α2b having the amino acid sequence designated as SEQ ID No.1, and
