titleOfInvention | IMPROVED METHODS FOR THE FORMATION OF DISULPHIDE LINKS |
abstract | 1. A method for forming a disulfide bond, including: causing or enabling the attachment of a first (poly)peptide/protein to a second (poly)peptide/protein, where said attachment is caused by the formation of a disulfide bond between the first cysteine residue contained in the specified first (poly)peptide /protein, and the second cysteine residue contained in the specified second (poly)peptide/protein, where one of the specified (poly)peptides/proteins contains an amino acid having a pI above 8, which positively affects the reactivity of at least one of the specified cysteine leftovers. ! 2. The method of claim 1, wherein said amino acid having a pI greater than 8 is contained in said first (poly)peptide/protein. ! 3. The method of claim 1, wherein said amino acid having a pI greater than 8 is contained in said second (poly)peptide/protein. ! 4. The method of claim 2 or 3, wherein said amino acid having a pI greater than 8 is not present at the corresponding amino acid position in said wild-type (poly)peptide/protein. ! 5. The method of claim 4, wherein said amino acid having a pI greater than 8 has been artificially introduced. ! 6. The method of claim 1, wherein said amino acid having a pI greater than 8 is contained within ten amino acids adjacent to said cysteine residue in the same (poly)peptide/protein. ! 7. The method of claim 6, wherein said amino acid having a pI greater than 8 is contained within five amino acids adjacent to said cysteine residue in the same (poly)peptide/protein. ! 8. The method of claim 1, wherein said first cysteine residue is present at or near the N-terminus or C-terminus of said first (poly)peptide/protein. ! 9. The method according to claim 8, where the specified first cysteine |
