titleOfInvention METHOD OF REPROGRAMMING SOMATIC CELLS
abstract 1. A purified preparation of isolated pluripotent mammalian cells, characterized in that the cells (a) express endogenous Oct4 and Nanog; (b) being injected with SCID to mice, they differentiate into tissue cells having characteristics of endoderm, mesoderm and ectoderm; (c) do not express the exogenously introduced pluripotency gene or a selective marker functionally linked to the endogenous pluripotency gene; (d) are reprogrammed somatic mammalian cells. ! 2. The purified cell preparation according to claim 1, characterized in that at least 50% of the cells are resistant to DNA demethylation. ! 3. The purified cell preparation according to claim 1, characterized in that the cells survive under conditions in which the expression of DNA methyltransferase I is reduced by at least 50%. ! 4. The purified cell preparation according to claim 1, characterized in that the cells are not genetically modified. ! 5. The purified cell preparation according to claim 1, characterized in that the cells contain at least one gene modification. ! 6. The purified cell preparation according to claim 1, characterized in that the cells genetically correspond to a donor of said somatic cells or a donor of a cell precursor of said somatic cells, said donor being an individual in need of cell therapy. ! 7. A method for generating a reprogrammed somatic cell, comprising using a somatic cell that contains one or more exogenously introduced genes that facilitate reprogramming the indicated cell into a pluripotent state, said cell being reprogrammed into a pluripotent state in which the cells are (i) resistant
