titleOfInvention | VIRAL VECTOR AND ITS APPLICATION |
abstract | 1. A viral vector comprising a Semlica forest virus replicon (SFV), wherein said replicon includes (i) a nucleotide sequence encoding an SFV replicase enzyme with mutations P718T and R649H in the nsp2 subunit, (ii) a polynucleotide comprising a selection gene, and (iii) polynucleotide comprising a nucleotide sequence encoding a heterologous target product. ! 2. The viral vector according to claim 1, comprising SEQ ID NO: 1 and SEQ ID NO: 2.! 3. The viral vector according to claim 1, including:! a) a polynucleotide comprising a nucleotide sequence encoding a heterologous target product, the expression of which is regulated by the first subgenomic SFV promoter (SG1); and! b) a polynucleotide comprising a selection gene, the expression of which is regulated by the second subgenomic SFV promoter (SG2). ! 4. The viral vector of claim 3, wherein said first and second subgenomic SFV promoters are identical. ! 5. The viral vector of claim 3, wherein said first and second subgenomic SFV promoters are different. ! 6. The viral vector according to claim 1, comprising, after the subgenomic promoter, a construct comprising a polynucleotide comprising a selection gene and a polynucleotide comprising a nucleotide sequence encoding a heterologous target product fused in phase. ! 7. The viral vector of claim 6, wherein said polynucleotide comprising a selection gene is fused in phase with a polynucleotide comprising a nucleotide sequence encoding a heterologous target product via a polynucleotide linker. ! 8. The viral vector of claim 7, wherein said linker is a polynucleotide comprising a nucleotide sequence encoding a site of post-translational (auto) proteolytic cleavage |
