titleOfInvention METHOD AND KIT FOR DETECTING A MICRO-ORGANISM
abstract 1. A method for detecting a living cell of a microorganism in a test sample, which includes the following stages:! a) adding a cross-linking agent capable of cross-linking DNA under irradiation with light having a wavelength of 350-700 nm to this test sample; ! b) irradiating the test sample to which the crosslinking agent has been added with light having a wavelength of 350-700 nm; ! c) removing the crosslinking agent contained in the light irradiated test sample; ! d) adding the medium to the test sample, from which the cross-linking agent has been removed, and incubating this test sample; ! e) adding again a cross-linking agent capable of cross-linking DNA under irradiation with light having a wavelength of 350-700 nm to the incubated test sample; ! f) irradiating the test sample to which the crosslinking agent is added with light having a wavelength of 350-700 nm; ! g) extracting DNA from this test sample and amplifying the target region of the extracted DNA using a nucleic acid amplification method; and ! h) analysis of the amplified product. ! 2. The method according to claim 1, wherein the amplified product is analyzed on the basis of a calibration curve that is generated using a standard sample of the microorganism and shows the relationship between the amount of this microorganism and the amplified product. ! 3. The method of claim 1, wherein the nucleic acid amplification method is a PCR, LAMP, SDA, LCR method, or a DNA microarray method. ! 4. The method of claim 2, wherein the nucleic acid amplification method is a PCR, LAMP, SDA, LCR method, or a DNA microarray method. ! 5. The method of claim 3, wherein the PCR method is performed using real-time PCR and both PCR and analysis of the amplified product are performed. ! 6. The method of claim 4, wherein the PCR method is performed
