titleOfInvention | MICROBIOLOGICAL METHOD FOR PRODUCING 3-HYDROXYI-BASIC ACID |
abstract | 1. A cell genetically modified compared to its wild type in such a way that it synthesizes more than 3-hydroxyisobutyric acid or polyhydroxyalkanoates based on 3-hydroxyisobutyric acid. ! 2. The cell according to claim 1, wherein the synthesis of 3-hydroxyisobutyric acid or polyhydroxyalkanoates based on 3-hydroxyisobutyric acid occurs with methyl malonic semi-aldehyde as the starting material. ! 3. The cell according to claim 2, wherein the cell synthesizes 3-hydroxyisobutyric acid or polyhydroxyalkanoates based on 3-hydroxyisobutyric acid with succinyl coenzyme A as an intermediate. ! 4. The cell according to claim 3, wherein the cell has an increased activity of the enzyme E1 compared to its wild type, which catalyzes the conversion of succinyl coenzyme A to methylmalonyl coenzyme A.! 5. The cell according to claim 4, wherein the enzyme E1 is methylmalonyl coenzyme A mutase (EC 5.4.99.2). ! 6. A cell according to one of claims 3 to 5, wherein the cell has increased compared to its wild type activity of at least one of the following enzymes from E2 to E4:! enzyme E2, catalyzing the conversion of methyl malonyl coenzyme A to methyl malonate; ! an E3 enzyme catalyzing the conversion of methyl malonate to methyl malonic semi-aldehyde; ! an E4 enzyme that catalyzes the conversion of methyl malonic semi-aldehyde to 3-hydroxyisobutyrate; ! 7. The cell according to claim 6, and the enzymes are as follows:! E2 methylmalonyl coenzyme A-hydrolase (EC 3.1.2.17),! E3 aldehyde dehydrogenase (EC 1.2.1.3) or aldehyde oxidase (EC 1.2.3.1) and! E4 3-hydroxyisobutyrate dehydrogenase (EC 1.1.1.31) or 3-hydroxyacyl coenzyme A-dehydrogenase (EC 1.1.1.35). ! 8. One cell |
