A polypeptide containing the human IGF-1 precursor protein, in which the cleavage of the E-peptide from IGF-1 by a protease is reduced as a result of modification of the precursor protein, and in which one or more of the following residues in the precursor protein is deleted by fission or modified by mutation: G1, P2, E3, R36 and R37. ! 2.
