titleOfInvention | METHOD FOR PRODUCING Sialylated Oligosaccharides |
abstract | 1. The method of production of oligosaccharides containing at least one sialic acid residue, which are here referred to as sialylated oligosaccharides, comprising the step of cultivating a microorganism in a culture medium possibly containing an exogenous precursor, said microorganism containing heterologous genes encoding CMP-Neu5Ac synthetase , sialic acid synthase, GlcNAc-6-phosphate-2-epimerase and sialyltransferase, and endogenous genes encoding sialic aldolase were delegated or inactivated nd acid (NanA) and ManNac-kinase (NanK), wherein said microorganism is capable of producing internally activated sialic acid. ! 2. The method of claim 1, wherein the heterologous sialyltransferase gene is selected from the genes for α-2,3-sialyltransferase, α-2,3- and α-2,8-sialyltransferase (cstll) and α-2,6-sialyltransferase. ! 3. The method according to claim 1, where the heterologous gene of CMP-Neu5Ac synthetase is neuA, the heterologous gene of sialic acid synthase is neuB, and the heterologous gene of GlcMac-6-phosphate-2-epimerase is neuC. ! 4. The method according to claim 3, where the neuA, neuB and neuC genes are isolated from bacterial strains containing a sialylated structure in their cell wall, such as, for example, C. jejuni strain ATCC accession number 43438.! 5. The method according to claim 1, where the genes nanT, nanA, nanK and nanЕ are delegated or inactivated (nanKЕАТ-). ! 6. The method according to claim 1, where the specified microorganism further encodes a protein that promotes the incorporation of lactose into the cells, and does not have enzymes that metabolize lactose. ! 7. The method according to claim 1, where the specified microorganism is a strain of E. coli, which is lacY + (β-galactoside permease), lacZ- (β-galactosidase) and, possibly, melA- (α-galactosidase). ! 8. The method of |
