titleOfInvention | METHOD FOR PRODUCING USEFUL METABOLITES |
abstract | 1. A method of producing a useful metabolite by fermenting a mixture of a primary carbon source and a secondary carbon source using bacteria modified so that the streams for utilization of the primary carbon source, such as carbohydrates or glycerin, and a secondary carbon source, such as ethanol, in said bacterium separated, including culturing the bacteria in a nutrient medium and isolating the metabolite from the culture fluid, characterized in that the bacterium has enhanced mutant expression alcohol dehydrogenase resistant to aerobic inactivation and capable of producing a useful metabolite selected from the group consisting of glutamate, proline, threonine, lysine, aspartate, asparagine, methionine, serine, valine, leucine, isoleucine, cysteine, arginine and pyrimidines, the fermentation of a mixture of a primary carbon source, such as carbohydrates or glycerin, and a secondary carbon source, such as ethanol, and is further modified so that the flows of carbohydrates or glycerin and ethanol are separated by attenuating SPRESSO gene (s) encoding (-their) pyruvate kinase. ! 2. The method according to claim 1, characterized in that the genes encoding pyruvate kinase include the genes pykF and pykA. ! 3. The method according to claim 1, characterized in that said expression of pykF or / and pykA gene (s) is attenuated by inactivation of pykF and / or pykA gene (s) in the bacterial chromosome. ! 4. The method according to claim 1, characterized in that the bacterium is further modified so that the expression of genes encoding proteins of a non-phosphotransferase transport system, such as xylE, galP, fucP, araE or lacY, is enhanced. ! 5. The method according to claim 1, characterized in that the decree |
