titleOfInvention | METHODS FOR CLEANING PROTEINS CONTAINING THE Fc AREA |
abstract | 1. A method for purifying a protein having an Fc region from an initial liquid containing said protein and one or more impurities, comprising the following steps:! protein adsorption on an Fc binding substance; ! washing the Fc binding agent with a buffer solution containing a salt of a divalent cation to remove one or more impurities; ! and isolating the protein from the Fc binding agent in an elution solution. ! 2. The method according to claim 1, characterized in that one or more impurities are selected from the group consisting of: varieties of reading introns (IRT), varieties with a low content of disulfide bonds (UDB) and varieties with low molecular weight (LMW). ! 3. The method according to claim 2, characterized in that one or more impurities are IRT. ! 4. The method according to claim 1, characterized in that the protein is an antibody. ! 5. The method according to claim 4, characterized in that the antibody is selected from the group consisting of: a fused construct containing the antibody, mouse antibodies, chimeric antibodies, humanized antibodies and human antibodies. ! 6. The method according to claim 4, characterized in that the antibody is an anti-IL-13 antibody. ! 7. The method according to claim 4, characterized in that the antibody binds an antigen selected from the group consisting of: CD3, CD52, VEGF, EGFR, CD33, CD20, HER-2, TNFα, CD25, RSV, IgE, gp IIb / IIIa, CD11a and integrin α4. ! 8. The method according to claim 1, characterized in that the protein containing the Fc region is obtained by recombinant DNA technology. ! 9. The method according to claim 1, characterized in that the protein containing the Fc region is obtained by recombinant DNA technology in a Chinese hamster ovary cell (CHO). ! 10. The method according to claim 1, characterized in that the Fc binding substance contains one or more of Protein A and Protein G.! 11. Method p |
