titleOfInvention | METHOD FOR PRODUCING ANTIVIRAL VACCINE IN SUSPENSION LINES OF ANIMAL EMBRYONIC STEM CELLS |
abstract | 1. The method of replication of a virus that is selected from the group consisting of adenoviruses, hepatadaviruses, herpes viruses, orthomyxoviruses, papovaviruses, paramyxoviruses, picornaviruses, poxviruses, reoviruses and retroviruses, in the bird embryonic stem cells of the ЕВх® stage, including: a) the proliferation of these EBx cells in culture vessels, in suspension, in serum-free medium No. 1; ! b) infection of said cells with a selected virus at a cell density of at least about 1.5 million cells / ml; ! c) adding serum-free medium No. 2 to the cell culture shortly before infection, simultaneously with infection or immediately after infection; and! d) further culturing said infected cells in order to create conditions for virus replication; and! e) optionally collecting said virus. ! 2. The method according to claim 1, whereby a continuously stirred bioreactor tank is used as a culture vessel. ! 3. The method according to claim 1, whereby serum-free medium No. 1 and serum-free medium No. 2 are one and the same medium. ! 4. The method according to claim 1, according to which serum-free medium No. 1 and serum-free medium No. 2 have a different composition. ! 5. The method according to claim 1, according to which from 0.25 to 10 volumes of serum-free medium No. 2 was added to serum-free medium No. 1. ! 6. The method according to claim 1, whereby serum-free medium No. 1 and / or serum-free medium No. 2 is supplemented with at least one ingredient selected from the group consisting of amino acids, lipids, fatty acids, cholesterol, carbohydrates, protein hydrolysates |
