titleOfInvention Method of oncoovarian acid-soluble alpha-2-globulin preparing
abstract Usage: in medical biochemistry for the isolation of onco-ovarian acid-soluble alpha-2-globulin from tissues necessary for the development of specific immunodiagnostics. The purpose of the invention: obtaining a new tumor-associated acid-soluble antigen. SUMMARY OF GETTING: Tumor and metastatic tissue extract of primary ovarian adenocarcinoma is heated in a boiling water bath for 5-20 minutes at 95 ° C. The heated extract is treated with an equal volume of 5-15% trichloroacetic acid. The insoluble fraction is precipitated by centrifugation. An acid-soluble fraction is fractionated with ammonium sulfate. Dissolve the precipitate in a minimal volume of distilled water and gel filter it on a Sephadex G-100 column. Precipitated with ammonium sulfate fractions containing GAG. Dissolve the precipitate in distilled water, followed by gel filtration on a Sephadex G-200 column in volatile 0.05-0.2 M ammonium acetate buffer (pH 5.0-7.0} and fraction lyophilization containing the target product. Positive effect: the invention allows, under ordinary laboratory conditions, using available reagents to obtain a new onco-ovarian f / yal acid-soluble human protein.
