abstract | Process for the preparation of Compound 1: comprising the steps of: i) providing 2-bromo-3-methylpyridine (Compound 2) and 3- (t-butoxycarbonyl) phenylboronic acid (Compound 3), ii) Cross coupling of Compound 2 and Compound 3 in biphasic a mixture consisting of water, a first organic solvent, a first base, and a transition metal catalyst to give compound 4, iii) oxidation of compound 4 to give compound 5, and iv) adding an amino group to position 6 of the pyridyl residue at to give compound 6, c) reacting compound 6 with compound 7 in a second organic solvent in the presence of a second base to give compound 8, vi) de-esterifying compound 8 in a biphasic mixture consisting of water, a third organic solvent. and first acids to give compound 9, and iii) suspending or dissolving compound 9 in a suitable solvent over an effective period of time to give compound 1. The application further comprises 32 claims. |
