titleOfInvention | NEW PROCESS OF SYNTHESIS (7-METHOXY-1-naphthyl) ACETONITRIL AND APPLICATION IN AGOMELATIN SYNTHESIS |
abstract | Process for the industrial synthesis of a compound of formula (I) characterized in that 7-methoxy-1-tetralone of formula (IV) is reacted with cyanoacetic acid of formula (V): under conditions of elimination of the water formed, in the presence of a catalytic amount of a compound of formula (VI) : wherein R and R ', the same or different, each represent a linear or branched (C3-C10) alkyl group, an unsubstituted or substituted aryl group, or an unsubstituted or substituted linear or branched (C1-C6) arylalkyl group, in order to after filtration and washing with basic solution, 7 (methoxy-3,4-dihydro-1-naphthalenyl) acetonitrile of formula (VII) is obtained: the compound of formula (VII) is reacted with a hydrogenation catalyst in the presence of an allyl derivative to, after filtration and evaporation of the solvent gave the compound of formula (I), the compound of formula (I) is isolated after recrystallization in the form of a solid, with the understanding that: - aryl means a phenyl, naphthyl or biphenyl group, - the term "substituted" refers to the formulas "aryl" and "arylalkyl" meaning that the aromatic moiety of these groups may be substituted by 1 to 3 of the same or different groups selected from linear or branched (C1-C6) alkyl, hydroxy and linear or branched (C1-C6) alkoxy, -under "allyl derivative" means a complete molecule containing 3 to 10 carbon atoms and may have another 1 to 5 oxygen atoms and, containing at least one motif-CH2-CH = CH2. The application contains 10 more claims. |
