abstract | The present invention relates to an analytic method for the specific highly-sensitive determination of steroid hormones in biological matrix, by solid-phase micro extraction, derivatization and detection. According to the invention, in a flask, provided with a septum and a magnetic stirrer, which contains the serum separated from blood, a polyacrylate fibre of 85 μ m is introduced through the septum, then NaCl is added for decreasing the estrogen solubility, the extraction is performed at 25...45°C for 100...120 min in thermostated bath, the fibre is withdrawn and the analytes are derivatized with N-methyl-N-trimethylsilyl-triflouro-acetamide and the silylated hormone derivatives are simultaneously determined by capillary gas chromatography coupled with mass spectroscopy. |
