abstract |
The invention relates to a bioprocess for the preparation of 7-amino-3-deacetyl-cephalosporanic acid and 7-aminocephalosporanic acid, adipic acid or one or more of its salts and esters, which have the capacity to be assimilated and used by Penicillium chrysogenum strain to produce adipoyl-6-amino-penicillanic acid followed by transformation of a strain of Penicillium chrysogenum with a gene coding for a expandase enzyme derived from Cephalosporium acremonium, which will in turn convert adipoyl -6-amino-penicillanic acid in adipoyl adipoyl 7-amino-deacetoxycephalosporanic acid, transforming a strain of Penicillium chrysogenum which produces isopenicillin N with a gene encoding an hydroxylase enzyme derived preferably from Cephalosporium acremonium which will convert in situ adipoyl-7-amino-deacetoxycephalosporanic acid in adipoyl 7-amino-deacetylcephalosporanic acid, which will undergo further enzymatic transformation to obtain 7-amino-3-deacetyl-cephalosporanic acid and 7-aminocephalosporanic acid. |
