abstract Prepn. of tumour necrosis factor (TNF) from mixts. with other proteins comprises adsorption onto a hydrophobic substance (A) or anion-exchange resin of uniform particle size, then eluting. (A) is esp. controlled pore glass or polystyrene beads (pref. eluted with ethylene glycol) and the ion-exchanger is esp. quat. ammonium substd. crosslinked polystyrene. Homogeneous TNF is new; esp. it is free of lymphotoxin and has activity about 10 million units/mg of protein. Also new are mutant forms of TNF having one or more amino acids substd., deleted or inserted; DNA coding for these mutant forms and cells transformed with this DNA.
