abstract | A method of fabrication of an immunosensor for detection of influenza virus, comprising: rinsing gold electrodes with ethanol and putting them into a 10 mM ethanol solution of 1,6-hexanedithiol (HDT) for around 20 h, at room temperature; rinsing the electrodes with ethanol and then deionized water, turning them upside down, and application of 10 muIota of colloidal gold solution (GCP) to them for 18 h at 4°C; in the next stage, rinsing the electrodes with 0.1 M PBS buffer (145 nM NaCI, 2.7 mM KCI, 8.4 mM NaH2PO4, and 1.6 mM Na2HPO4) and application of 10 muIota of anti-M antibodies at the concentration of 28 mug/ml in 0.1 M PBS buffer to the electrodes for 20 h at 4°C; in the final stage, application of 10 muIota of 0.5% (w/v) BSA in 0.1 M 2 h at 4°C; rinsing the electrodes with PBS buffer and storing at 4°C until use (not longer than 1 day). Also, the use of the immunosensor for detection of M protein and influenza virus was restricted. |
