abstract Referred to variants and isoforms of bispecific antibodies, Emicizumab with reduced biological activity. A variant of an antibody CHARACTERIZED BECAUSE it comprises a variable region comprising the amino acid sequence SISPSGQSTYYRREVKG (SEQ ID NO: 2), wherein: (a) the amino acid residue R in the 12th position from the N-terminal side of the sequence; or (b) amino acid residues YYR between the 10th and 12th position from the N-terminal side of the sequence; and the variable region is excised at the deletion site. It also refers to an isoform of a bispecific antibody CHARACTERIZED BECAUSE it comprises a first heavy chain and a second heavy chain, where disulfide bridges are formed in the following positions: (1a) between the cysteine at position 144 according to the EU numbering of the first heavy chain and the cysteine of position 200 according to the EU numbering of the second heavy chain; and (1b) between the cysteine at position 200 according to the EU numbering of the first heavy chain and the cysteine at position 144 according to the EU numbering of the second heavy chain; or where disulfide bridges are formed in the following positions: (2a) between the cysteine at position 226 according to the EU numbering of the first heavy chain and the cysteine at position 229 according to the EU numbering of the second chain heavy; and (2b) between the cysteine at position 229 according to the EU numbering of the first heavy chain and the cysteine at position 226 according to the EU numbering of the second heavy chain. These antibody variants and isoforms have reduced FVIII mimicry activity compared to Emicizumab. Furthermore, it relates to detection methods and analysis methods for the variants and isoforms of antibodies.
