abstract This invention is in the field of Insulin-like growth factors 1 (IGF-1) modifications. It relates to modified IGF-1 polypeptides and modified IGF-1 precursor polypeptides wherein the cleavage of E-peptide is prevented. In particular, the present invention provides a polypeptide comprising a human IGF-1 protein variant having at least 90% identity to SEQ ID NO: 1, wherein the amino acid glycine at position 42 is mutated to serine. The invention also relates to the use of such polypeptides for treating muscle diseases and disorders.
