abstract |
Provided is a process for production of a purified recombinant human arylsulphatase A (rhASA) in a continuous cell culture system, the process comprising: i) continuously culturing a mammalian cell capable of secreting rhASA into liquid culture medium; ii) concentrating, purifying and formulating the rhASA by a purification process comprising one or more steps of affinity chromatography and/or ion exchange chromatography; wherein the rhASA produced has a specified amino acid sequence or its variant, wherein the process of (ii) comprises a polishing step including a passive step, wherein the rhASA passes through a cation exchange resin or membrane without binding thereto, and an active step, wherein the rhASA is detained within an anion exchange membrane or resin and subsequently eluted from said anion exchange membrane or resin; wherein the cation exchange resin or membrane and the anion exchange membrane or resin are coupled or connected in a series; and wherein the continuous culturing is for a period of at least one week. |
