abstract | Disclosed is a method for indirectly coupling a small molecule ligand to a molecule to be labelled with the ligand, the method comprising the steps of: attaching at least one small molecule ligand which incorporates an N-hydroxy succinimide derivative to a scaffold molecule (such as dextran derivatives); and subsequently contacting, in a single mixture, the scaffold molecule (with the attached at least one small molecule ligand); the molecule to be labelled (which is an enzyme, an antibody or an antigen-binding fragment or variant of an antibody etc.); and a thiol-generator (which is typically 2-iminothiolane (also known as Traut's reagent); wherein the thiol-generator forms thiol groups in situ on the molecule to be labelled; the scaffold molecule having at least one group which is reactive towards the thiol group formed on the molecule to be labelled, so as to form a bond between the scaffold molecule and the molecule to be labelled, thereby indirectly coupling the small molecule ligand to the molecule to be labelled; and wherein the small molecule ligand is typically selected from the group consisting of: fluorophores; chromophores; biotin; avidin; metal ion chelators; photoreactive groups; iodinatable moieties; photosensitisers; quenchers; peptides; and low molecular weight drugs. |
