abstract | A method for detecting whether a microbe specific nucleic acid sequence is present in a biological sample, comprising the steps of: 1. Providing an aqueous composition in pre-mixed form; wherein the composition consists essentially of a mix containing components sufficient for a polymerase chain reaction (PCR) which include: - Forward and reverse amplification primers specific for PCR amplification of the nucleic acid sequence; - A PCR polymerase enzyme; - One or more buffers; - A plurality of nucleotides sufficient for PCR amplification of the nucleotide sequence; - A detectable probe for detection of the nucleic acid sequence after PCR amplification; and - The mix of combined components remains substantially stable for at least two weeks when maintained at a temperature of between 18 to 27 degrees celcius. 2. Extracting nucleic acid from a biological sample; 3. Adding the extracted nucleic acid to the pre-mixed aqueous composition; 4. Subjecting the pre-mixed composition and added nucleic acid to PCR amplification under conditions where the nucleic acid sequence for detection may be amplified; and 5. Assaying the composition to detect the presence of the amplified nucleic acid sequence. |
