abstract |
A process for recovering the heparin binding protein, vascular endothelial growth factor (VEGF), from a prokaryotic cell culture. The process comprises the steps: (a) isolating the VEGF from the periplasm of the prokaryotic cell culture; (b) denaturing the VEGF in a first buffered solution that comprises a chaotropic agent and reducing agent; (c) incubating the denatured VEGF in a second buffer solution that comprises a chaotropic agent and a sulphated polyanionic agent for a time sufficient to refold the VEGF; (d) recovering the refolded VEGF, wherein there is a 2-5 fold increase in refolded VEGF compared to incubating with no sulphated polyanionic agent. The polyanionic agent can be dextran sulphate or sodium sulphate and the VEGF can be VEGF165. |
